چکیده

The objective of this study was to distinguish between males and females of jojoba (Simmondisia chinensis) using Random Amplified Polymorphic DNA (RAPD) markers in order to detect any DNA variation. A callus was produced from both male and female jojoba leaves, using MS medium with BA 0.5 and 2 to 4, D 2.5 mg/l respectively. Genomic DNA of jojoba plant was extracted using CTAB method. The concentration of DNA ranging between 100 to 150 μg with DNA purity ranging from 1.01 to 1.19. About 0.7 g of male and female plant leaves, and calli were used. Eight different random primers were evaluated for their usefulness in detecting DNA variation between male and female of jojoba. This involved optimization of RAPD reaction condition including, DNA template, Taq polymerase, and primers. Five primers showed no amplification and represent a mean of distinguishing between male and female. One primer (C5) showed amplification and represent a mean of distinguishing between the two sexes from the fresh sample tissue; the callus results show no significant difference. The results demonstrated the feasibility of using RAPD-PCR in distinguishing between jojoba sexes.

Failure of PCR-RAPD technique to differentiate sex in Mahisefied (Rutilus frisii kutum) from the South Caspian Sea

In order to identify the sex marker in Mahisefied, Rutilus frisii kutum, samples from 5 male and 5 female fish were collected from the south Caspian Sea. Polymerase chain reaction random amplified polymorphic DNA (PCR-RAPD) was performed using 124 primer sets. All bands were numbered using 1 and 0 scores corresponding to the presence or absence of bands, respectively and data were analyzed usin...

A high genetic diversity revealed between commercial rose cultivars by RAPD-PCR technique

Genetic diversity between fifteen commercial cultivars of rose were evaluated, using Ten decamer primers (A-J). All primers detected polymorphism among the cultivars. In total, 126 bands were produced, 73 of which were polymorphic. Primers E and F produced the highest, while primer H produced the lowest number of bands. The percentage of polymorphic bands ranged from 37% to 81% with an average ...

Molecular differentiation of sheep and cattle isolates of Fasciola hepatica using RAPD-PCR

Understanding genetic structure and status of genetic variation of Fasciola hepatica isolates from different hosts, has important implications on epidemiology and effective control of fasciolosis. Random amplified polymorphic DNA (RAPD-PCR) was used to study the genetic variation of F. hepatica in sheep and cattle. DNA was extracted from adult helminthes removed from livers of each infected ani...

A comparative analysis of ISSR and RAPD markers for studying genetic diversity in Iranian pistachio cultivars

Pistachio is one of the most important horticultural products of Iran. The best way to get the maximum yield is to have genetically pure and monotonous gardens. Thereby, study of genetic variation and providing genetic identifications, make this possible to have homogenous gardens with high performance genotypes. In the present study, genetic diversity of 19 Iranian pistachio cultivars was asse...
